转录是基因表达的第一步,因此非常重要。本课题组致力于构建独特的纳米芯片,阐明转录机制和细胞命运调控模式,开发新的精确控制细胞命运的手段。
研究成果展示
In synthetic biology, the control of gene expression requires a multistep processing of biological signals. The key steps are sensing the environment, computing information and outputting products. To achieve such functions, the laborious, combinational networking of enzymes and substrate-genes is required, and to resolve problems, sophisticated design automation tools have been introduced. However, the complexity of genetic circuits remains low because it is difficult to completely avoid crosstalk between the circuits. To overcome these problems, we have combined a synthetic biology approach (EMBO J. 2010) and a biophysical approach (Nature 2015, Mol Cell 2015, 2018, 2019), making a unique nano-chip by integrating an actuator (enzyme) and sensors onto a DNA origami-based nanochip (Nature Nanotechnology 2018). This gene nanochip allows us to rationally design the chemical reaction and precise gene expression control in vitro and in vivo. Our approach to component integration on a nanochip may provide a basis for large-scale, integrated genetic circuits.
